I'm surprised no one has mentioned John Wayne as Rooster Cogburn in "True Grit"! His gunfight against "Lucky" Ned Pepper, the Original Mexican Bob and two others was a true classic.

Great...simply great.

Yep, that was one of the two I was going to mention. Love John Wayne movies. "I intend to shoot you in one minute Ned or hang at Judge Parker's convenience, which will it be?" This is my number one classic western fight scene.

Second, would be the gunfight in the saloon at the end of Unforgiven w/Clint Eastwood. "You just shot an unarmed man! Well, he should arm himself if he's going to decorate his saloon w/my friend.... I've killed most everything that walked or crawled at one time or another and I'm here to kill you Little Bill."

Quotes probably aren't perfect, I'm going from memory.
